Simvastatin

Short-acting statins(PO) are given at night to reduce peak cholesterol synthesis in the early morning.They are generally absorbed and extracted by the liver,their site of action &  are subjected to widespread presystemic metabolism via cyt P450 &  glucuronidation pathways. Simvastatin is an inactive lacton prodrug; it is predominantly metabolized in the liver to form the corresponding active beta-hydroxy fatty acid.
Clinical Uses:
·         Atrovastatin lowers serum cholesterol in patients with homozygous familial hypercholesterolaemia.
·         In severe drug resistant dyslipidaemia (e.g. heterozygous familial hypercholesterolaemia).Ezetimide is combined with statin treatment.
